چکیده

In plants, Lewisa type N-glycans may be involved in cell-to-cell communication and recognition. N-glycoproteins harboring Lewisa glycotopes are mainly found in plasma membranes and cell walls. Some can be also involved in cell wall synthesis or the loosening process, and subsequently in cell elongation. In order to determine the potential role(s) of the alpha4-fucosylation during vegetative development, transgenic tobacco plants overexpressing a human Lewis fucosyltransferase (hFUT3), which transfers a fucose residue in a alpha(1,4)-linkage on complex glycans, have been developed. The heterologous enzyme hFUT3 was strongly expressed and fully functional in transgenic tobacco. Transgenic plants showed a delay in growth linked to a reduction of internode length. Furthermore, transgenic seedling roots were significantly shorter than wild-type roots and the length of their epidermis cells was reduced. Strikingly, root growth was completely and specifically restored following gibberellin treatment. Etiolated hypocotyls of hFUT3 overexpressors were also more sensitive to exogenous gibberellin. Furthermore, paclobutrazol, an inhibitor of gibberellin synthesis, induced a similar effect on control and transgenic dark-grown hypocotyls suggesting that gibberellin biosynthesis was probably not altered in seedlings overexpressing hFUT3. Thus, alpha4-fucosylation could act as a possible modulator of conformation and/or functioning of N-glycoproteins involved in the gibberellin-dependent elongation process.

Expression and antimicrobial activity analysis of dermaseptin B1 recombinant peptides in tobacco transgenic plants

Recently, new molecular breeding and genetic engineering approaches have emerged to overcome the limitations of conventional breeding methods in generating disease-resistance transgenic plants. The use of antimicrobial peptides (AMPs) to produce transgenic plants resistant to a wide range of plant pathogens has achieved great success. Among huge number of AMPs, Dermaseptin B1 (DrsB1), an antimi...

Biotechnological Reduction of Tobacco (Nicotiana Tabacum L.) Toxicity

BACKGROUND: Nicotiana tobacco contains large amounts of alkaloid nicotine. Tobacco plant is used for smoking and causes many health problems since it is high in nicotine which is one of the widely-recognized toxic compounds with serious side effects for different body organs. Reducing nicotine content of this plant is a way to reduce its health hazards in cigarette smokers. Utilizing the new ...

Assessment of salt tolerance in transgenic tobacco (Nicotiana tobacum L.) plants expressing the AUX gene

Transformation of plants using Agrabacterium rhizogenes may affect secondary metabolite production as well as morphological changes. In this study, T-DNA from Ri plasmid in A. rhizogenes carrying pRi15834-PRT35S-GUS was introduced into tobacco leaf segments to initiate development of transformed hairy roots. Plant regeneration from transgenic roots used MS medium, and plants regenerated fro...

Gibberellin homeostasis in tobacco is regulated by gibberellin metabolism genes with different gibberellin sensitivity.

Gibberellins are phytohormones that regulate growth and development of plants. Gibberellin homeostasis is maintained by feedback regulation of gibberellin metabolism genes. To understand this regulation, we manipulated the gibberellin pathway in tobacco and studied its effects on the morphological phenotype, gibberellin levels and the expression of endogenous gibberellin metabolism genes. The o...

Responses of Transgenic Tobacco (Nicotiana plambaginifolia) Over-Expressing P5CS Gene Underin vitroSalt Stress

Salinity is a major limiting factor for plant growth and development. To evaluate the impact of P5CS gene expression under in vitro salt stress condition, transgenic tobacco (Nicotiana plumbaginifolia) carrying P5CS gene and non-transgenic plants were treated with 0, 100, 150, 200 or 250 mM NaCl for 28 days. Proline content, lipid peroxidation and the activity of some antioxidant enzymes after ...
